Understanding Stability Studies for Dietary Supplements

Ara Testing Labs has been receiving questions recently about regulatory expectations and when stability studies are needed. This article shares insight that will help you understand both topics on a deeper level.

Much of the confusion comes from how expiration dates, “best‑by” dates, and manufacture dates are interpreted.

The Importance of the Expiration or “Best By” Date

The key point is simple and often overlooked:

If a dietary supplement carries an expiration or best‑by date, the manufacturer must have scientific stability data that demonstrates the product meets all label claims through that date.

This requirement is tied to the FDA’s dietary supplement cGMP rule in 21 CFR Part 111, which requires products to meet their identity, purity, strength, and composition specifications throughout their shelf life.

Instead, many brands choose to use a Manufacture Date (MFG). A manufacture date is a factual statement showing when the product was produced, and it does not require long‑term stability data. FDA does not prohibit the use of MFG dates. Under the Dietary Supplement Health and Education Act of 1994 (DSHEA), manufacturers are responsible for ensuring products are not adulterated or misbranded, but DSHEA does not require expiration dating on supplements.

June 2025 Draft Q1 Stability Guidance

In June 2025, the FDA released an updated draft guidance titled Q1 Stability Testing of Drug Substances and Drug Products (Draft Guidance, June 2025). This document consolidates and updates all prior ICH stability guidance (Q1A–Q1E and related documents) into a single harmonized resource.

This document outlines expectations for:

  • Stability study design
  • Data evaluation
  • Packaging considerations
  • Degradation modeling
  • Statistical approaches

Even though this guidance is written for drugs, supplement manufacturers that choose to label products with expiration dates increasingly use these principles to support scientific justification.

What Stability Studies Typically Include

Study Types

    • Real-time studies
    • Accelerated studies

Common Storage Conditions

    • 25 °C / 60% RH (long‑term, temperate)
    • 30 °C / 65–75% RH (intermediate)
    • 40 °C / 75% RH (accelerated)

We have tested many products and have seen that some formulations, particularly enzymes and probiotics, degrade rapidly when exposed to elevated temperature and humidity. For these categories, accelerated conditions may not provide meaningful or predictive data, and ambient real‑time studies are often the most appropriate option.

Typical Timepoints
  • 0, 1, 3, 6, 9, 12 months
  • 18 –24 or even 36 months for real‑time, long‑term studies
  • Accelerated studies are often completed within six months
    • The industry practice uses a 4:1 predictive correlation (for example, 3 months accelerated ≈ 12 months real time)

Testing Performed
  • Potency: HPLC, UPLC, LC‑MS, UV/Vis
  • Microbiology: APC, yeast and mold, indicator organisms, pathogen absence (risk‑based)
  • Physical attributes: Color, odor, texture
  • Other specific product tests, as required.

In short: If you want an expiration date on the label, you need stability data to back it up.
